Storage/Stability
dPGA is highly stable and can be stored at 4 °C for up to 24 months.
Stock Preparation
Under sterile conditions, dilute the dPGA solution to a 1 mg/ml stock solution with sterile ddH20.
A sterile 1 mg/ml stock solution can be stored at 4 °C long term (up to 2 years).
Dendrimer Coating Protocol
When using dPGA as an surface attachment factor, the optimal conditions must be determined for each cell line and application. In general, the following steps can be used.
- Under sterile conditions, dilute the dPGA 1mg/ml stock solution to a final concentration of 50µg/mlcoating (1:20 dilution) in sterile 1XPBS.
The final concentration of the coating solution may vary from 10-100 μg/ml. We recommend using a concentration of dPGA that is the same as the concentration of poly-lysine in similar protocols. - Coat the surface of the culture vessel with the dPGA working solution. Add enough solution to cover the culture surface.
- Incubate for at least 1 hr at 37°C (we recommend in a CO2 incubator).
- Remove the solution by aspiration and rinse 3X with sterile 1XPBS.
- After the last rinse, seed the cells or coat with a second layer of Laminin or Matrigel (see Step 6 below).
Coating a second layer with laminin or Matrigel - After step 5 (above), the dPGA coated substrate may be coated with laminin or Matrigel. Proceed with your usual coating protocol. Incubate the plates for 1-2 h at 37 °C or overnight at 4 °C.
